Morphological characteristics of leukemia cells in acute myeloblastic leukemia with t(8;21)(q22;q22): possible predictability of t(8;21)

چکیده
The laboratory systems for chromosomal analysis or the detection of fusion genes are generally not available in Indonesia. Therefore, bone marrow (BM) morphological analysis should be developed and applied to get an accurate diagnosis. In this study the BM smears of eight (8) cases of acute myeloblastic leukemia (AML) which had already been known to have t(8;21)(q22;q22), were morphologically evaluated in order to find out the characteristics, which might be used to predict t(8;21)(q22;q22) or the presence of AML1-ETO(MTG8) fusion gene. All of the cases belonged to AML-M2. The morphological characteristics, indicative of t(8;21) were pink colored cytoplasm in mature neutrophil (75%), neutrophilic myelocytes or metamyelocytes without granules or with scarce granules (2.3%), eosinophilia (eosinophilic myelocytes and metamyelocytes) (above 5%), myelocytes with abundant granules 8.5%, and low percentage of type I blasts (below 10%). These characteristics were not observed in AML-M2 cases without t(8;21) or AML1ETO(MTG8). The myelocytes with abundant granules have not been described so far, while other characteristics were in line with the findings of Nakamura et al (Leukemia 1997;11:651-55). Distinctive immunophenotypic features of t(8;21)(q22;q22) acute myeloblastic leukemia in children.
Thirty cases of newly diagnosed pediatric acute myeloblastic leukemia (AML) with French-American-British (FAB) M2 morphology were analyzed with cytogenetics and a comprehensive panel of monoclonal antibodies reactive with lymphoid-, natural killer (NK)-cell-, and myeloid-associated antigens. The t(8;21)(q22;q22), or t(8;21;V)(q22;q22;V), translocation was identified in 16 of the 30 cases. Cases...
متن کاملExpression of CD7 antigen precludes t(8;21)(q22;q22) chromosome aberration in acute myeloblastic leukemia.
The report by Ball et all showed the significance of the lymphoid markers, CD2 and CD19 antigens, expressed by subsets of acute myeloblastic leukemia (AML) in correlation with chromosomal aberrations and prognosis. The report disregarded CD7 antigen as lymphoid marker, probably because the relatively high incidence of the expression of CD7 antigen in AML cases, compared with CD2 or CD19 antigen...
متن کاملGeographic Heterogeneity of the AML1-ETO Fusion Gene in Iranian Patients with Acute Myeloid Leukemia
Background: The human AML1 gene, located on chromosome 21, can be fused to the AML1- eight-twenty-one (ETO) oncoprotein on chromosome eight, resulting in a t(8;21)(q22;q22) translocation. Acute myeloid leukemia (AML) associated with this translocation is considered a distinct AML with a favorable prognosis. Due to the various incidences of the translocation, which is associated with geographic ...
متن کاملA rare cytogenetic presentation of acute myeloid leukemia (AML-M2).
Acute myeloid leukemia (AML) with t(8;21)(q22;q22) generating the AML1/ETO fusion gene on 8q22 is a distinct type of AML t(8;21) category (WHO)/AML-M2 (FAB), generally associated with a favourable prognosis. Variant additional chromosomal abnormalities are frequently reported. We report three adult cases of this category with unusual karyotype. Bone marrow cytogenetics of case no. 1: 45,X,-Y, t...
متن کاملDuplication of der(21)t(8;21)(q22;q22) in acute myeloid leukemia.
The t(8;21)(q22;q22) results in the formation of RUNX1/ RUNX1T1 (alias AML1/ETO) and RUNX1T1/RUNX1 fusion genes on der(8)t(8;21)(q22;q22) and der(21)t(8;21)(q22;q 22), respectively. An 18-year-old woman was diagnosed as having acute myeloid leukemia (AML) M2, as her bone marrow was infiltrated with 42.2% myeloblasts. Auer rods were found in myeloblasts and metamyelocytes (Picture A, B, arrows)....
